Strep-Tactin™ purification buffer set

Strep-Tactin™ Buffer Set contains all buffers necessary for purification of Strep-tag™II or Twin-Strep-tag™ fusion proteins with Strep-Tactin™ as well as for subsequent regeneration of the resin. Strep-Tactin™XT Buffer Set comprises the following buffers:
• 10x Buffer W, 100 ml
• 10x Buffer BXT, 25 ml
• 10x Buffer XT-R, 250 ml 10x Buffer W is a 10x concentrated wash buffer for Strep-Tactin™ and Strep-Tactin™XT resins. It removes unbound molecules from the affinity chromatography matrix during Strep-tag™II and Twin-Strep-tag™ fusion protein purification. 10x Buffer E is a 10x concentrated Strep-Tactin™ elution buffer. The contained desthiobiotin is a modified form of biotin that displaces Strep-tag™II and Twin-Strep-tag™ fusion proteins from the modified biotin-binding pocket of Strep-Tactin™ in a competitive manner, resulting in a mild elution. Please note that 10x Buffer E is not applicable for elution from Strep-Tactin™XT resins. 10x Buffer R is a 10x concentrated Strep-Tactin™ regeneration buffer containing HABA. HABA (2-[4-hydroxy-benzeneazo]benzoic acid) displaces desthiobiotin from the modified biotin-binding pocket of Strep-Tactin™ after the elution of Strep-tag™II or Twin-Strep-tag™ fusion proteins. The yellow buffer turns red upon binding of HABA to Strep-Tactin™, clearly indicating the regeneration process and activity status of the column.
Please note that the Strep-Tactin™ Buffer Set is not applicable for Strep-Tactin™XT.
