A chemically defined, protein-free, phenol red-free buffer for complexing plasmid DNA with transfection reagent for standard transfection applications.

Gibco Viral-Plex Complexation Buffer is a ready-to-use AAV production buffer and complexation reagent for gene therapy, formulated to efficiently complex plasmid DNA with lipid-based transfection reagents for viral vector manufacturing. This chemically defined, serum- and protein-free buffer provides an optimal environment for forming stable DNA/lipid complexes, enabling efficient transient transfection of Viral Production Cells 2.0 (VPC2.0). As a key component of the AAV-MAX Production System, Viral-Plex Complexation Buffer streamlines the triple-transfection workflow for high-titer AAV production.

Features of the Viral-Plex Complexation Buffer

  • Chemically defined, animal origin–free formulation—protein- and phenol red–free buffer provides a controlled environment for consistent DNA–lipid complex formation
  • Optimized for AAV workflows—designed for use with the AAV-MAX Production System to enable efficient, helper-free triple-transfection in suspension culture, streamlining gene therapy vector production
  • High-titer, scalable production—supports AAV yields greater than 5 x 1010 vg/mL (unpurified) in suspension culture; seamlessly scales from shake flasks to bioreactors for large-scale viral vector manufacturing
  • Ready-to-use solution—supplied as a 1X formulation (e.g., 100 mL bottle) requiring no preparation; mix with plasmid DNA for immediate transfection use
  • Consistent performance—a stable, pre-formulated buffer ensures reproducible transfection efficiency and batch-to-batch consistency in viral vector manufacturing

Applications

  • Gene therapy—ideal for the production of adeno-associated viral (AAV) vectors for gene therapy research and development
  • Cell therapy—supports the generation of adeno-associated viral (AAV) vectors for cell therapy applications, facilitating seamless transition from bench to clinic
  • Research and development—provides a reliable and scalable solution for viral vector production in various research settings

Available in RUO or CTS

Offered as both a Research Use Only (RUO) version and a GMP-compliant Cell Therapy Systems (CTS) version (Cat. No. A5145403) to support translational and clinical-scale applications.
