In 1957, Jeffries et al. developed a medium to demonstrate DNase activity of microorganisms that possess the extracellular enzyme deoxyribonuclease1. The DNase enzyme catalyzes the cleavage of phosphodiester linkages of DNA leading to its degradation. Organisms producing DNase enzymes thus show clear zones around the colonies. DNase Test Agar is used to identify various bacteria like staphylococci, enteric gram-negative bacilli and pseudomonads2-5.
DNase Test Agar is used to detect the deoxyribonuclease (DNase) activity in microorganisms.
- Easy-to-interpret: After flooding the plate with hydrochloric acid (1N):
- Positive test – a clear zone around the growth line and the rest of the plate remains opaque.
- Recommended for identification of variety of bacteria like staphylococci, enteric gram-negative bacilli and pseudomonads2-5.
DNase Test Agar contains casein and soy peptone that supply nitrogen, amino acids and peptides necessary for bacterial growth. Sodium chloride provides essential electrolytes and maintains osmotic equilibrium.
DNase producing organisms depolymerize DNA into mononucleotides and oligonucleotides when 1N HCl is added to DNase Test Agar after incubation. HCl reacts with polymerized DNA in the medium to provide free nucleic acids and a cloudy precipitate. In areas where DNA is depolymerized, a clear zone is formed around the DNase producing colonies.
