Extracted from Crotalus adamanteus venom.

Phosphodiesterase I is widely used in studying nucleic acid structure and sequence. It hydrolyzes 5'-mononucleotides from 3'-hydroxy-terminated DNA and RNA. Phosphodiesterase I cleaves ADP-ribosylated proteins at the pyrophosphate linkages to yield phosphoribosyl-AMP.
