- Recognizes human PG II
- Pepsinogen I and II are precursors of pepsin, preproduced by gastric mucosa and released into gastric lumen and peripheral circulation
- Advanced inflammation of gastric mucosa and its progression toward atrophic gastritis in corpus is associated with a change in serum biomarkers pepsinogen I and II
- PG II (Pepsinogen II, progastricsin, PGC, pepsinogen C) is an aspartic protease expressed in gastric chief cells and novel marker of Type 2 cells with advantages over current markers used to identify Type II cells in developing lung
- Involved in proteolysis and peptidolysis
Process:
- Antibody specific for PG II pre-coated onto microwells
- PG II protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific for PG II is added to detect captured PG II protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
