Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous MMP-2

  • Recognizes human MMP-2
  • MMP-2 (Type IV collagenase) cleaves collagen Types IV, V, VII and X and gelatin Type I
    • Activation of MMP-2 secretion requires Ras signaling pathway
    • Ubiquitinous metalloproteinase involved in diverse functions (remodeling of vasculature, angiogenesis, tissue repair, tumor invasion, inflammation and atherosclerotic plaque rupture)
    • Contributes to myocardial oxidative stress by regulating GSK3beta activity

Process:

  • Antibody specific for MMP-2 pre-coated onto microwells
  • MMP-2 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for MMP-2 is added to detect captured MMP-2 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
