Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous Mammaglobin A

  • Recognizes human Mammaglobin A
  • Mammaglobin A (SCGB2A2) is a member of secretoglobin superfamily
    • Expressed in normal mammary gland epithelium and over-expressed in primary and metastatic human breast cancers, makes it a specific molecular marker and potential therapeutic target for breast cancer
    • Secreted Mammaglobin protein detected in sera of breast cancer patients, not in sera patients with other tumor types
    • Serum marker for breast cancer

Process:

  • Antibody specific for Mammaglobin A pre-coated onto microwells
  • Mammaglobin A protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for Mammaglobin A is added to detect captured Mammaglobin A protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
