- Recognizes human INS
- Insulin is a peptide hormone, produced by beta cells of pancreas and is central to regulating carbohydrates and fat metabolism in the body
- Participates in glucose utilization, protein synthesis and formation and storage of neutral lipids
- Insulin synthesizes as precursor molecule, proinsulin, which is processed prior to secretion
- A and B peptides are joined together by a disulfide bond to form insulin, while central portion of precursor molecule is cleaved and released as C-peptide
- Defects in insulin results in Type 1 diabetes mellitus
Process:
- Antibody specific for INS pre-coated onto microwells
- INS protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for INS is added to detect captured INS protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
