- IL-12 (p70) is a 70 kDa heterodimer composed of 35 kDa subunit (IL-12A p35) and 40 kDa subunit (IL-12B p40)
- IL-12 involved indifferentiation of native T cells into Th1 cells
- T cell-stimulating factor can stimulate growth and function of T cells
- Plays role in activities of natural killer cells and T lymphocytes

Process:
- Antibody specific for IL12 p70 pre-coated onto microwells
- IL12 p70 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for IL12 p70 is added to detect captured IL12 p70 protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
