Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous IL1-beta

  • Recognizes human IL1-beta
  • Interleukin-1 is a pro-inflammatory cytokine with multiple biological effects
    • IL-1 gene family encodes IL-1α, IL1-beta, IL-1RN
    • IL1-beta produced by blood monocytes and tissue macrophages
      • Implicated in mediating both acute and chronic inflammation
    • Involved in cellular activities (cell proliferation, differentiation, apoptosis)
    • Key mediator of carcinogenesis that characterizes host-environment interactions

Process:

  • Antibody specific for IL1-beta pre-coated onto microwells
  • IL1-beta protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for IL1-beta is added to detect captured IL1-beta protein
  • For signal development, Streptavidin-HRP is added followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
