Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous HSP90

  • Recognizes human HSP90 protein
  • HSP90 encoded by HSP90AA1 is a constituitively and ubiquitously expressed molecular chaperone is crucial for stability and function of proteins
    • Provides chaperoning activity for client proteins, implication in tumor malignancy
    • Mainly residues in cytosol, can be released into extracellular space
    • Level of plasma HSP90 is positively correlated with tumor malignancy in clinical cancer patients, promising diagnostic marker for tumor malignancy in clinical application

Process:

  • Antibody specific for HSP90 pre-coated onto microwells
  • HSP90 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for HSP90 is added to detect captured HSP90 protein
  • For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
