Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous CYB5R3

  • Recognizes human CYB5R3
  • Kit for testing serum, plasma and cell lysates
  • CYB5R3 (DIA1, B5R) belongs to flavoprotein pyridine nucleotide cytochrome reductase family
    • Two naturally occurring isoforms
      • Isoform 1: anchored to cytoplasmic side of endoplasmic reticulum membrane and mitochrondrion outer membrane
      • Isoform 2: soluble form found in red blood cells

Process:

  • Antibody specific CYB5R3 pre-coated onto microwells
  • CYB5R3 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody specific for CYB5R3 is added to detect captured CYB5R3 protein
  • For signal detection, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
