Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous CD147

  • Recognizes human CD147
  • CD147 (Basigin, BSG or EMMPRIN) is a transmembrane glycoprotein belonging to immunoglobulin superfamily
    • Important in numerous physiological and pathological phenomena
    • Involved in reproduction, neural function, inflammation and tumor invasion
    • CD147 expressed at high levels at retinal pigment epithelium and neonatal blood-brain barrier on tumor cells and activated T cells
    • Can stimulate adjacent fibroblasts to produce matrix metalloproteinases
    • Effective marker to predict invasion and prognosis in various cancers
    • Soluble CD147 in body fluids associated with systemic sclerosis, hepatocellular carcinoma, ankylosing spondylitis and coronary diseases

Process:

  • Antibody specific for CD147 pre-coated onto microwells
  • CD147 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for human CD147 is added to detect captured human CD147 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
